Currently, Illumina 5-base supports only PCR prep
The epigenetic methylation of cytosine bases in DNA can have a dramatic effect on gene expression. Bisulfite sequencing is the most canonical method for detecting epigenetic methylation patterns at single-base resolution through converting all unmethylated cytosines to thymine. These libraries inform methylation but have low base diversity which precludes accurate genomic interrogations. Novel Illumina 5-base assay converts methylated cytosine to thymine, retaining high complexity, and is capable of simultaneous variant calling and DNA methylation levels detection.
